LAMP Master Mixes
Loop mediated isothermal amplification (LAMP) is a amplification technique that has played an increasingly important role in point-of-care (POC) diagnostics. It offers several advantages over qPCR and other amplification techniques in terms of sample-to-answer time, sensitivity, specificity, cost, robustness, and accessibility, making it ideal for field-deployable diagnostics in resource-limited regions.
High-Sensitivity LAMP Master Mixes for Rapid POCT Development
- Inhibitor-tolerant isothermal mix designed for direct isothermal amplification from crude samples.
- Very fast reaction kinetics – ideal for point of care testing.
- Fully optimized, just add custom primers and probes.
LAMP master mixes enabling highly sensitive detection of target DNA or RNA. Meridian has also developed sample specific inhibitor tolerant LAMP mixes, designed to be used for direct amplification from crude lysates or inhibitor-rich samples, with the ability to tolerate even higher levels of inhibitors present in samples. In addition, these mixes can be used as wet mixes or dried by lyophilization or air-drying for room temperature shipping and storage, extended shelf-life, and increased flexibility in sample volume, making them ideal for POCT development where minimal sample processing and fast turn-around times are essential.

FAQs
How is isothermal amplification different from qPCR?
The main difference between loop mediated isothermal amplification and qPCR is that in loop mediated isothermal amplification, the amplification is achieved using a single constant temperature, while in the qPCR, amplification requires cycling between multiple temperatures on a thermocycler. Instead of melting DNA strands apart at high temperatures, loop mediated isothermal amplification uses polymerases with high strand displacement activity, like Bst DNA polymerases. As these polymerases do not need multiple temperatures, they can amplify a target in less than an hour, and in some cases in as little time as 10 minutes.
Can LAMP be used for a multiplex assay?
Yes, it is possible to design a multiplex LAMP test using different primers and probes, however they can be difficult to design and optimize as each sequence requires three sets of primers.
What is the performance difference for DNA when using the DNA-specific product versus the DNA/RNA product?
We do not see a performance difference between the two mixes, which is why we call the Lyo-Ready Direct RT-LAMP Blood mix – Lyo-Ready Direct RNA/DNA LAMP Blood mix, as it can be used for both.
Are reaction conditions the same for lyophilized and liquid mixes?
Yes, the LAMP and RT-LAMP master mixes can be used as a liquid mix or lyophilized and stored for up to 24 months without needing to change the reaction conditions and this drying/ambient temperature storage will not affect the sensitivity of the POCT.
